While Dean and Cas did not have much of a relationship in season 1, they quicky formed an alliance in season 2, also known as Twopernatural. However, Castiel ended up making poor decisions leading to Dean mocking him behind his back and making him a target. Soon after, Dean makes his distrust of Castiel known, while Sam still cared about Cas, leading to Sam's iconic line: "Dean, I voted for you tonight." Dean and Castiel were eliminated in a double elimination.

In season 3, Dean and Cas slowly start to become friends, and maybe even more than friends.

A majority of Destiel fans are very vocal in their beliefs that Dean and Castiel are romantically drawn to each other. Dean is often referred to as Castiel's favorite contestant. It is often joked that their first meeting was "love at first sarcastic comment."

Many believe that Dean does not treat Castiel like a friend or a brother but more like someone he loves romantically. This is referring to the many moments on the show where there is romantic tension between the two. Characters in the show have made jokes about the two of them dating. Sam has often been caught in the middle of a "Destiel" moment, and has awkwardly had to cough to get the two to snap out of it.

On AO3, Destiel is the most written ship for Dean and Castiel. However, it is not the most written ship in the Supernatural (TV) tag nor the most written ship on AO3. On April 5th 2023, Destiel had like 27 fics written, which is an unimpressive feat.
